I would consider the attack on Fort Sumter to be the first battle of the Civil War, however, First Bull Run (or First Manassas as referred to by the Confederates) was the first major engagement where organized armies from both sides faced off.
At this point in time many, many people believed that the rebellion was little more than saber rattling, and that if it did actually come to shooting, the Union would have it wrapped up in a few weeks. At Bull Run people were coming in droves from miles around to see the "battle", congressmen, senators, and Washington D.C.'s wealthy elite were packing picnics, bringing the kids and family pets, and making a Sunday outing of it. The battle proved to be an embarrassing Union defeat ending in Union forces being routed, with soldiers dropping guns and abandoning equipment, to run back to Washington as fast as they could. It was the first time many people considered that there really was a rebellion and that they really were at war. The conclusion of First Bull Run brought the stark reality that the largest, bloodiest, and most deadly battle in American history had just occurred.

It was Fort Sumter, South Carolina on the night of April 12, 1861.
When President Lincoln planned to send supplies to Fort Sumter, he alerted the state in advance, in an attempt to avoid hostilities. South Carolina, however, feared a trick; the commander of the fort, Robert Anderson, was asked to surrender immediately. Anderson offered to surrender, but only after he had exhausted his supplies. His offer was rejected, and on April 12, the Civil War began with shots fired on the fort. Fort Sumter eventually was surrendered to South Carolina.

It could be said that the first battle of the U.S. Civil War was fought at Fort Sumter near Charleston, South Carolina. Because South Carolina had already seceded, President Abraham Lincoln was lucky enough to have a Southern fort that was still under Northern influence. Being low on supplies, Lincoln could either have ignored it & have it be sent under Confederate power, or send supplies & alert the South. As history would have it, Lincoln sends supplies, & the Confederate immediately thinks that it is an act of aggresion. They would eventually use warships and attack Union carriers that would go near Fort Sumter. It was essentially a Confederate victory with no deaths.
The first pitched battle was Bull Run (Manassas), which brought the country to a little bit of reality - that the war would not be something short and glorious.
The first shots were fired three months earlier at Fort Sumter (signalling the start of hostilities) and the Confederates celebrated this rather one-sided event as a victory, though it could not really be classified as a battle.
There had also been various skirmishes elsewhere, which the press wanted to dramatise, and this led to Irvin McDowall's miserable experience at Bull Run, when he already knew that his army was not ready for combat.
