That is a difficult question to answer, because while American women did not get the right to vote in federal elections till 1920, a number of states and territories had already given women the right to vote-- the first was the Wyoming territory in 1869. Certain cities also had given women the right to vote for particular officials-- in Boston, for example, women could vote for school committee members beginning in 1873.
Some sources say that Lydia Taft was the first woman in America to vote, because she was allowed to cast her ballot in her local town meeting in Uxbridge MA in 1756; it should be noted that this was permitted because her wealthy husband had died and her son was a minor, so the townspeople agreed that the family should in fact have a vote at town elections and gave Lydia that right. But in most cases, wives and widows were not granted voting rights, nor would they be until many more years had passed. But other sources note that Louisa Ann Swain was the first woman to vote, because she cast a ballot in the general election in Wyoming in 1870. She was 70 years old when she achieved this milestone.
The first women to vote was Margaret Brent.
No one knows for sure. But the first woman to vote was Mrs.Marie Ruoff Byrum, wife of Morris Bryum, daughter-in-law of Lacy Byrum.
