If you can prove that it was self-defence and didn't break any laws concerning the possession and use of the knife, yes. However if they die, you could be tried for manslaughter.

The United States does not legally recognize any diference between men and woman in any way, self defence or not. So no, it is legal. Wether that person was capable to cause injury to yourself/others enought to provoke an act of self defence is a different matter.
