Of course you can! That is, providing you have the necessary equipment to do so. For example, if you are playing Guitar Hero on Wii, you need two guitars and two Wii remotes. For other consoles, I am unsure, but it is in my understanding that you can buy wireless guitars that would work perfectly fine providing you have the correct console to work with the guitar.
Rockband for the Wii can be bought with or without the instruments. If you buy Rockband with the instruments, it should come with one guitar, one drum set, two drumsticks, and a microphone.
