Not directly, but most Job Applications do ask if the applicant is a US citizen/national or legal resident. This is a roundabout way for an employer to know an applicant's alien status, but the official reason is to establish that the applicant has a legal right to work. Of course, if the applicant provides false papers, that is a different matter.

There's really no need to. When they fill out an application, they're required to answer whether or not they're a US citizen, whether or not they have a legal right to work in the US, and if they are able to bring documented proof of their status. So, they're asked indirectly, anyhow.
