They work with gaseous uranium hexafluoride, if you have a long line of hundreds of centrifuges and pass the gas through them, you can gradually spin off some of the heavier U-238 resulting in a main stream with a greater percentage of U-235. This method has been used in Europe for many years and uses much less power than the older gaseous diffusion method which has been continued in the US, but I think that is now changing to centrifuges. This is what the State Dept is so concerned about happening in Iran. If you have enough centrifuges you can get nearly pure U-235 out which of course can be used in weapons.
