Lifters can take time to pump up at first when starting....but worn main bearings can knock also
well it could bet a bent push rod or a lifter could be bad
well a couple things could be wrong depending on what year. if it is a hydraulic lifter then the oil pressure might not be high enough for the oil to reach the lifter causing a knock. another problem could be that your rod and main bearings could be worn out causing a knock and low oil pressure.

If it's just one or two windows it could be the motors ate sticking, take the door panel off, locate the motor, and jive it a good squirt of WD40, also any moving parts.

The modulator valve is located on the LH side of the tranny and has a vacuum line going to. After replacing you have to set the modulator pressure by connecting a gauge to the tranny, removing the cover from the modulator valve and turning adjustment bar underneath. With out pictures its hard to show how the gauge is connected. To be quite honest if you really have done the tests required to diagnose the modulator than setting the pressure should be the easy. Vacuum lifters are usually rented. To purchase a vacuum lifter, one could look for a manufacturer or ask a rental company if they can order one in to sell.

You could take off a rocker and try to push down on the pushrod. You'd be able to feel the spring in a hydraulic lifter, but not on a solid lifter.
