The Ohlone had their culture forcibly taken from them and were denied the right to their language or their religion by the Christian missionaries and Spanish colonizers; thus, a case could be made that they were probably not happy. On the other hand, since they were subjugated, nobody asked about their happiness, and perhaps eventually, they became accustomed to their new way of life, since they did not have a choice.
In fairness, it should be pointed out that the missionaries of that era sincerely believed they were doing the natives a service; they did not regard the conquest of the native peoples as a bad thing. Rather, they believed it would improve the condition of the Ohlone if they were converted to Christianity. And by living at the mission, they would then have access to the dominant (Spanish/Catholic) culture.
