in one way yes, in another, no. when you reach the area called the "vault" it turns out to just be a portal to a massive boss call the "destroyer" and the storyline was set for you to kill it, not to really ever make it into the vault. either way, it has great gameplay and it's really fun.
borderlands ends by you killing the destroyer and not getting into the vault. the armory ends by you killing general knoxx and blowing up the armory

You get to enter the area called 'The Vault' where you have to kill a large 'Boss' monster which guards the entrance, upon killing the Boss the vault locks down for another 200 years.

There are 4 vault hunters that are featured in borderland's 2's new game. The game was released in September 2012. In the new release of the game the vault hunters are tasked with killing Jack and returning peace to Pandora.
Borderlands 2 follows on from the original Borderlands, in which a guardian 'Angel' guides a team of 4 'Vault Hunters' to find and open a mysterious Vault. The Vault turns out to be a prison for an alien monstrosity, which the Vault Hunters defeat. Unknown to them, 'Angel' is a signal being broadcast from a Hyperion company satellite, setting the scene for Borderlands 2. In Borderlands 2, Hyperion's CEO Handsome Jack is beseiging the game's main planet of Pandora to wipe any opposition out, in order to reopen the vault. New Vault Hunters arrive, only one (the player) survives Jack's attack and joins the resistance, which is lead by the original team of Vault Hunters, met over a large portion of the game. Though guided again by Angel, it turns out she is working for Jack, betraying the resistance - but after this she offers genuine help in order to help defeat him. It turns out that she is human, not an AI as presumed, and that Jack is her father. She ends up dying, though Jack manages to kill one member of the original hunters, and kidnap another. Ultimately Jack opens the vault, gaining control of a guardian beast inside, a warrior with almost limitless power. The player is able to defeat the warrior before it reaches full strength, and kill Jack in a final showdown. Throughout the game the player meets many eccentric characters, who help, hinder, and occasionally annoy.

You can not go back in and out of Vault 101. After you first leave the Vault the only time you can go back in is for the quest "Trouble On The Homefront." but once you beat that side quest you are not allowed back in Vault 101 ever.
