I just bought a pair of suede boots, and even though i havent spilled nail polish on them, here's what i would do: Use some nail polish remover and a piece of tissue, and kind of blot the spot. Then rub gently and it should come out.

First you want to clean the shoes, with either special soap made for shoes or a very very little amount of rubbing alcohol because to much will dry the leather. After that you take a smooth cloth, add shoe polish, spritz a little water on it and then rub thoroughly into the shoe. Once you have covered all over the shoe you want to buff it. You can buff it two different ways. Either using a brush constantly to remove the excess polish and keep brushing until shoe is cleaned and shined. Or you can use the brush once or twice and then use a shoe buff cloth and rub it left and right real quickly which buffs and cleans the shoe as well.
