It depends on which operating system you want to boot on your flash drive. I have been using an application called Bootsage to create a Windows 7 installer flash drive or a WinPE bootable environment, and it seems to work very well. It runs under Windows XP as well as Windows 7.

One good way is to get Ultimate Boot CD 4 Windows and use it to make a bootable USB. It will give you some good tools on that drive.

My Computer, right click on floppy drive-> Format... In the next windows choose Create a MS-DOS startup disk

It won't make any difference at all. I have non system disks in my drives all the time. It matters not.
To make your flash drive boot windows XP simply install windows to your flash drive to do this insert flash drive into USB port on PC or laptop. Insert windows operating disc and install to said drive for where you inserted the flash drive. Once the install is complete remove the flash drive and turn off PC or laptop insert flash drive and boot PC or laptop and go to your Bios and change the boot sequence to the flash drive well it worked for me
In Windows, press F8 during booting.. You will find a menu in which you will fine an option - "Safe Mode in DOS Prompt".. Enter in that menu, you will boot from DOS Prompt.. In Windows 98, you will find the "Boot from DOS prompt".. You can select that option to boot from DOS..
