Just a thought. I have a Toyota Sienna also. The gas compartment is on the left side. If the gas compartment is even popped then there is a safety mechanism that will not let the door open. Your compartment might be closed, but you could be having a mechanical problem with the compartment that would cause the safety mechanism to come on. Simple, but could just be it.
There is a push button that can be pressed up or down (it is rectangular in shape) located below the air vent to the left of the steering column.
Just push the "MANUAL DOOR" switch at the vicinity of the dashboard at the left and above of the stereo ; it should lock-out the electrical and you can just open and close the door manually.
I have the same problem on my 05' Sienna. My mechanic showed me that it was the door hinge making the noise (soon your window will not open unless door is open). My mechanic said he has seen this before, but Toyota dealer has to fix it. They want $1K to fix, so we are living with it. Toyota is going down hill.
There is not a fuse that controls the air to your vents in your 2000 Toyota. The vents are controlled by blending doors. The doors open and close to direct air to the desired location.
My 2004 Honda Odyssey sliding door will not close all the way. I begin to close for 3 quarters of the way an then stops. I have to force it to close manuallt. Thanks

I would think the bolt sliding forward in the receiver would close the aperture.
If the sliding door will not close on the 2005 Saturn Relay, look for something that is stuck in the door latch area. Also check the latch to see if it has come out of adjustment. The pin or hook might need to be adjusted or tightened.
