You add the Transmission Fluid to where you check the level (I think it's on the passenger side when you look under the hood). remove the dip stick and add fluid with a narrow funnel. Check level when engine is running.
If there is no drain plug you will have to remove the pan to drain the fluid. This removes 4-5 quarts out of 12 or so in the transmission. You can also take it to a shop and have them do a fluid exchange where they pump out all the old fluid and replace it with new.
If it has one, you simply use the correct size socket and ratchet and turn counterclockwise.

The water pump on a Ford Econoline E250 van is changed by removing the accessories from the front of the engine, draining the coolant, and removing the radiator hoses. The pump can then be unbolted, removed, and a new pump put in its place.
