Depends why the light is on. At my shop I've reset the light with a scan tool and drove through a drive cycle and inspected the vehicle. Unless it's a major problem or a sensor is showing zero reading you can usually put 20-60 highway majority miles and pass. Most vehicles require multiple consecutive failed emission readings before triggering the check engine light.
Yes, I believe it will, Based on the NYS inspection site. The mechanic will advise you that the light is on. It is not included in the OBD II diagnostics which is sent to DMV. Check out the site and judge for yourself, or ask a mechanic you trust.

That will cause you to fail an emissions inspection. Anything which triggers the CEL will, because everything which triggers it will adversely impact the emissions system on that vehicle.

FYI, I had a mechanice replace the O2 sensor and the check engine light came back on again. I then changed the air filter and took out the Mass Air Flow sensor and cleaned it with electrical parts cleaner and put it back in. I also added a fuel injector cleaner to a full tank of gas. I reset the light by removing the negative cable to the battery for 30 seconds. This worked well enough for the car to pass a NYS inspection.
When it says so IM NOT KIDDING Actually, it expires at the end of the month that it is punched for. So, if it is punched MARCH, it expires the end of that month!!
the same sticker is used for trailer inspections, if it were used on a trailer the "t" would be punched out.
Expired inspection sticker IS a state charge, there are no local jurisdiction ordnances that equate to it.
