Open the rear door and lift to the full up position. Look at the top of the inside rear window and you will see a panel (molding) running across the top of the rear window. Gently pry it off. It is held in place with 4 metal spring clips. Be sure not to lose any of the clips and note what position they are in. The three clips to the right are similiar, the one on the left is longer.
Once you have removed the panel, in the center you will see the bulb holder with two wires running out of it. Rotate it counter-clockwise and remove.
You should now see the exposed bulb. It is a #921. Pull it straight out and replace. Regretfully the bulbs are sold in 'two packs' and this bulb is not used anywhere else on the Highlander.
Reinsert the bulb holder, rotate clockwise, and give it a gentle tug to ensure it's locked in. I recommend you have someone step on the brake pedal at this time to test the light before replacing the panel. If it lights up, replace the panel (molding) and you're done.

Open the rear hatch, there is a plastic panel that is at the top of the hatch which is long and thin.. It snaps out and the light is behind it. Careful though, the clips break.. I took it Toyota, I couldn't figure it out..
