In the engine compartment are 2 dip sticks. One going to the engine and one to the transmission. Pull out the one for the transmission and read what it says. Nearly all transmissions want the car running, warm and in park. The dip stick usually states this on the side.
If these requirements are on the side or nothing different from that, then pull it out clean it off and then reinsert it. Pull it out quickly and check the level.
