To clean a streak plate I use coke classic. In the red can, the diet version does not work as well. I believe it is the acid that takes away the powdered rock debris. It makes them quite white again. Mr clean magic eraser also takes away some of the heavy debris...but does not get them clean enough.

A contaminant is anything other than the organism(s) you were trying to grow. This question is a little like asking what plants are weeds and there is no 100% true answer. The purpose of a streak plate is to gain single isolated colonies. The most troublesome species to find growing on your streak plates are swarming bacteria as they spread and contaminate other colonies, pseudomonas sp. are an example. The contaminating bacteria could have come from the operative or from the room where the procedure was carried out.
The spread plate can become contaminated if you forget to sterilize your "hockey" stick or leave the plate open on a counter.

More ways than you can count. Forget to sterilize your 'hockey' stick, have a contaminated culture to begin with, leave the plate open on the bench, sneeze while spreading, have a air conditioner turn over while spreading, have a sloppy bench mate, etc.....
