The two subway lines that stop at Penn Station are the 1-2-3 (the red line) and the A-C-E (the blue line). The 1-2-3 stops at the east side of Penn Station (at 7th Avenue) and the A-C-E stops at the west side (at 8th Avenue).
Both of these subway lines will drop you off very close to 221 Cadman Plaza, so you can just take whichever one you see first.
