I would just walk. Even if you were coming from the far west end of Grand Street (Varick Street or 6th Avenue) it would only be about a mile. From the east end (by the East River) it would be 3/4 of a mile, and from Little Italy or the Bowery or Chrystie Street, it's only a few blocks.

Plus, there is no direct subway route.

To get there, walk west or east to Chrystie Street, then walk 2 blocks south on Chrystie to Canal Street, and the roadway to the Manhattan Bridge will be right there. You will be able to see the bridge from the corner of Chrystie and Canal.
