Apart from the obvious "it's not moving anymore", you can tell if it's truly dead because it will become stiff after a few hours when rigamortis has set in.
If your hamster is dead, it will be unresponsive to light, sounds, or touch. If you touch it, it may feel stiff, depending on how long it has been dead. If it is sick, but not dead, it may be more likely to jump and bite you if you touch it, so you may want to very gently touch it with something like a pencil eraser if you are not sure.
