You can block them by clicking on the "block [username]" link found on users profile pages. Twitter well then show you 2 buttons asking you to confirm that you really mean to block this person from following you.
You can get tweets from a User A that you're not following if another User B that you follow retweets User A's tweet.
To stop getting tweets by User A that have been retweeted by User B, unfollow User B.
You can also turn off retweets for a specific user if you don't like what they share. Select Turn off Retweets from the person icon drop down on a user's profile to stop seeing Tweets they've retweeted.

You will receive tweets on your homepage from everyone you follow. Unfollow them to not receive their tweets. To stop people seeing your tweets (except from those you approve) you can protect your tweets from the settings menu.

People won't be able to use the Retweet function on your tweets if your tweets are protected. This doesn't stop them copying and pasting your tweet and retweeting the old way though.
Go to settings and tick the box that says "Protect your tweets"
If you click on the "Follow" button, they will always know you are following them as you will appear in their "Followers" list. However, there's nothing to stop you from looking at their tweets (unless their profile is private) and just not clicking the "Follow" button.
To unprotect your tweets go to settings and uncheck the protect tweets box.
