I'll make some assupmtions here:
It's not a 3500HD truck. The HDs have a full floating rear axle that can be removed without pulling the wheels. You just unbolt the outside cap and pull it out.
If it's a truck, it is a standard duty either 1500 or 2500. You're going to have to pull the diff cover and partially disassemble the locker to get at the ends of the axles. There's a C clip at the end of the axle that has to be pulled out in order to release it. You have to put a little inward pressure on the axle in order to allow the clip to be pulled off. A good service manual should have pictures to help you.
If it's a car, the issues are the same as above.
