Need to know exactly what year and what size V6 in order to provide an answer.
Some don't have wires, the coils are mounted direct to the spark plugs. Some have a dual spark plug design where the plug mounted coil also sparks another spark plug on the other side of the engine. Removint these spark plug wires will still allow the engine to run but will eventually ruin the coils. And the third type will not run with out wires.

I WOULD SUSPECT THAT ONE OF THE PLUG WIRES IS NOT MAKING CONTACT OR IS NOW BROKEN.
the problem is your ignition coil pack going to your spark plugs
make sure you didnt put one of plug wires on the wrong plug if you did it will crank and run but it will run ruff/miss
the spark plug provides the spark necessary to ignite the fuel and air in the engine, which allows your engine to run. without spark plugs, your engine wouldn't start.
First raise the hood. Then locate the spark plug wires. DO ONE AT A TIME OR ELSE YOUR CAR WILL NOT RUN VERY WELL OR NOT AT ALL. Remove the first spark plug using a ratchet with a 6" extension (this prevents busted-up knuckles)and a 5/8" spark plug socket. Loosen and remove. When replacing with a new plug be careful how tight you get the plug, as you will want to do this again in 1 year. Oh, and by the way, you can apply antisieze compound to the spark plug threads; but please do so sparingly.
A spark plug, tork is something that sparks to run but the torque gives it extra power because it is a cadillac

it could be 1.3mm or 0.051inch although apparently ford re-gap there plugs to 1.0mm so cars can run on "supermarket fuel"
A miss on one cylinder is caused by defective spark plug, plug wire, burnt valve, or blown head gasket. Replace all plugs and plug wires. If the miss is still there run a compression test.
