If the pump is a modern pump with 2 inch Plumbing then 3-5 hours will filter the entire pool volume. To get an exact calc. you need to know the feet of head in the plumbing and compare it to a manufacturers pump curve chart but the above answer will probably be correct.

(btw: 45K gallon is huge).. check your pump/filter it should say how many gallons per minuate it can pump. figure out how many hours it would take to pump though 45K gallons (ie: 100 gal/min x 60 min/hr = 6000 gal = hr, 45k/6k = 7.5 hrs).. have it go though the pump twice a day...
