A car battery needs all 6 cells to work.
The hardest job a car battery has to do, is crank the engine over during starting. This draws hundreds of amps (depending on engine size).
If even one cell is down, the internal resistance will severely restrict the amperage delivered and the motor won't turn.
Surely a battery with a dead cell can appear to be live. The lights and radio may work, but dimly. How long this will continue, will depend on how many ancillaries are switched on and the original charge and capacity of the battery.
A battery that cannot start the car is next to useless.

If, by 'larger battery' you mean one with the samevoltage but higher capacity - there's no reason why not. It will simply take a little longer to charge it. I swapped my 3.6v 600mAh battery for a 3.6v 1000mAh battery a long time ago - and the phone still works perfectly well.
