It depends on what route you're taking. If you're taking the Trans-Canada, probably about 3 1/2-4 hours depending on whether you hit traffic in Winnipeg and assuming you do not stop.

About four and a half hours, depending on highway speed, and what route you take. From the Ontario border to Winnipeg on Highway No. 1 is about an hour and a half. From Winnipeg to Portage la Prairie is another hour. If you continue on to Brandon on Highway No. 1 it is another hour to Brandon and another hour or so to the Saskatchewan border. If you take Highway 16 from Portage (heading to Saskatoon rather than Regina) then it is three hours from Portage to the Saskatchewan border at Russell, so your entire trip will be an hour longer.
