Depends. The general rule of thumb is that a pet snake should be given 48 hours to digest their meal(s). If they are handled before that time is up, the snake may regurgitate the food. This is natural: in the wild, if a snake is attacked by a predator soon after eating a meal, they puke up the food in order to be able to escape quickly. If the snake is an adult, handled regularly, very docile, and used to the handler, it should be OK to handle it within 48 hours. This should never be attempted with a baby snake, as they are usually not used to being handled and will most likely vomit on the handler, which is not fun to clean up, trust me :)! I have a pet snake that allows me to handle him an hour after feeding without incident. So, it all depends on how tame and docile the snake is.

Only if the gloves aren't clean ! There's no telling what chemicals may be on them. On the other hand (no pun intended) if you have a particularly aggressive snake, that's not used to being handled, using gardening gloves will lessen the chance of the snakes teeth biting into your hand. Most snakes quickly become accustomed to being handled - then you can discard the gloves.
