You can use any mode.. Normally it can use from safe mode and normal mode..
When a program cannot run in DOS mode it means it's a Windows file. Unless you get a specific program (you'll have to look into it) you can't use any Windows programs.
Either remove it from Start>>Control Panel>>Add or Remove Programs(In Windows XP) Start/Windows Logo>>Control Panel>>Programs and Features>>Uninstall a program(In Windows Vista and Windows 7) If you are unable to remove it from there or you are unable to find the program in the list use the uninstaller for that program from Start/Windows Logo>>All Programs and then select the program and then uninstall If for some reasons like virus you are unable to remove a program by Control Panel method you can start your PC in Safe mode(For Safe mode restart PC and keep tapping F8) and then uninstall it.
