Yes, a bad coil will affect any combination or all of the cylinders - it is the source of all spark required for firing the spark plugs and making the engine run.

the 4.6L and 5.4L Ford V-8's and 6.8L V-10's have one coil pack for each cylinder or spark plug. You need to remove the 7mm hex head bolt and unplug the coil pack from the wiring harness, then remove the coil pack unit from the spark plug hole and do an ohm test that is the only way to test these.

Coil packs can be checked for resistance with an ohm meter. If the pack is breaking down due to load or heat problems then the only way to check is to change it out. If the Windstar is a two pack model then you can swap the packs to see if the problem follows the cylinders that the possibly bad coil pack are now controlling.
