Rear end gear ratios are matched to the engine/trans combo. A v8 will turn at a lower rpm than a v6 so it would be best to match the ratios so you do not lose low end power, a v6 will turn at a higher rpm than a v8 at the same speed.

There is really no advantage to a posi rear end except for traction, your steering might become more difficult though.

Another problem will be the drive shaft spline size, length and strength, in most cases the smaller engine will use a lighter duty shaft and the mounting at the diff will be different than the v6, along with u-joint size, spring mounting and shock mounting points, and brakes.
