HAHA i would say the best thing to do for that is to let the paint set up and cut the drip or sag off with a razor. sand flat. then touch up. While it is wet is a real trick. ive never seen it done :/
The tire itself may be bad, with a flat spot (no tread) or bulge. Also, a wheel that is badly out of balance can thump at some speeds.
