No, not necessarily. Most likely the drain has become rotten because of something acidic going into it. Vent pipes usually have a good flow of air in both directions. I would just change the bad waste pipe(s).

If it's leaking from the back of the water pump then most likely you should go ahead and replace it because its leaking from the weep hole which tells you basicly that your water pump is about to stop working its a way to warn you before your motor locks up

There is a weep hole on the underside of the water pump. If it is leaking out of that weep hole, it is defective and needs replacing. If the gasket is leaking which is unlikely if the pump is an OEM pump, you need to just go ahead and replace the pump anyway. If you go to all the trouble and labor to remove the pump in order to replace the gasket it would be foolish to not just go ahead and install a new water pump instead of reinstalling the old one, which may fail at any time.
