Your programs and your data in the computer are stored on the hard drive (with very few exceptions). RAM memory is the 'working' memory of the machine, it is volatile memory that is erased when the power is turned off, so removing or replacing RAM will not delete anything from your hard drive.
That said, you should always have a current backup of your important data and programs before you undertake any repairs or upgrades to your computer.
