The damage itself doesn't matter, Indestructable creatures can't be destroyed by damage. A 3/3 Indestructable creature could have 0, 1, 3, or 100 damage counters on it, and it would be fine, but if a -3/-3 effect is then used on it, it dies because its toughness has reached 0. The amount of damage on it at that point is irrelevant.

It means "destroy" effects don't work on it. Darksteel Colossus can't be shattered, disenchanted, wrath of God, Seal of doom, etc. However since it can be sacrificed, cards like Chainer's Edict still work. Also reducing it's stats would work, so Mutilate can kill Indestructible creatures. Damage or combat damage destroys creatures, so it wouldn't destroy an indestructible creature. However it does take the damage, but the damage doesn't kill it.
Deathtouch is a static ability, like trample or flying, that reads, "When this creature deals damage to another creature, destroy the creature that was dealt damage." For example, if you have a 1/1 creature with deathtouch blocking an 8/8 creature, then your creature will deal 1 damage to the attacking creature, and vice versa. Your creature will die because it was dealt enough damage to kill it, but your opponent's creature will die as well. This ability also works when you are attacking or using an ability that deals damage to a creature, such as Pestilence Demon.

Creatures that has first strike deals their combat damage before creatures without. So if a 2/2 creature with first strike blocks a 3/2 creature without first strike the 3/2 will die from the first strike damage and the 2/2 will not take any damage.
