If you import save data from Mass Effect 1, then whichever character lived through the first game will be in ME2. If you don't import save data and don't use Genesis, Kaiden will be in the game, but Ashley won't.
If you played Mass Effect 1 the second game imports your old game save, so it would depend on how you played Mass Effect 1 as to whether or not Kaiden is alive. If you did not play Mass Effect 1, then the game will default to the character with the opposite gender to Shepard. So Ashley will be there with a male Shepard, Kaiden for a female one. Depending on what happened in the previous 2 games, the characters are varied slightly. When starting a new game, if you chose for Kaiden to live, then Ashley won't be available as a character and same goes for if you choose to let Ashley survive. Available squadmates throughout the game are: Garrus Liara Kaiden (or Ashley). James EDI - She will become available later in the game. Tali'Zorah - She can only be a squadmate for a few missions and she will appear later on in the game. Shepard's allies are: Joker ('Jeff') Engineer Adams Admiral Hackett Luitenant Cortez Specialist Traynor Dr. Karin Chakwas Diana Allers - She is a reporter who will request interviews after certain missions.
