I can't answer re. all cars, but the Transmission Fluid dipstick in my Ford Taurus is as easy to get to as the oil dipstick. Since the transmission fluid also often needs checked frequently my guess is it is usually as easy.
Look under the hood, and locate the transmission dipstick. Grab the top, and pull the dipstick out of the dipstick tube.
if you know where the oil dipstick is then it should be the only other dipstick under the hood
If it has an automatic transmission, it should have a dipstick on the passenger side, under the hood, near the firewall. If it has a manual transmission, no dipstick.
There should be a dipstick under the hood. it should be labeld TRANSMISSION FLUID. its probably got a red cap on the dipstick.
To find the dipstick, look for a thin, plastic handle under the hood of the car. This is the transmission dipstick and can be used to check the fluid.

Assuming you have a automatic transmission the dipstick should be located under the hood, behind the engine, on the passenger side.

Assuming you have an automatic transmission the dipstick is located under the hood on the passenger side in back of the engine, close to the firewall.
The transmission in a 2016 Ram 1500 does not have a dipstick. The fluid is checked using a set of steps through a fill port on the side of the transmission. The dipstick on a 2500-5500 is under the hood.
Its under the hood towards the back. Check for a red colored dipstick handle.
