There are lots that will show you the difference between values in different cells. All you need to do is subtract one from the other, normally the lower one from the higher one. So if you wanted to find the difference between a value in A3 and a value in B3, where A3 had the higher value, then you would use the following formula:

=A3-B3

You can also do it in such a way where it doesn't matter which is the highest, by using the ABS function. Sometimes that is better as when you are setting up the spreadsheet, you won't always know which cells will have the higher values. If all you need to know is what is the difference no matter which is the higher one, then you would do it like this:

=ABS(A3-B3)

You would use a subtraction. If there is a time in each of two cells, say cell A2 and B2, then you would subtract the earlier time from the later time:

=B2-A2

There is no single answer to that. It would depend on what you mean by difference. You could mean difference in terms of two numbers. You could mean the difference between other data types like text, date and logical values. You could mean difference in size or colour or font. So there could be all sorts of formulas used.
