No. When you attempt to buy something on the computer that says you still have money, iTunes will log you in and update how much money you have in your account before the purchase is made. I know of cases where, due to a glitch on the iPhone (which has probably been patched by now), some users accidentally managed to spend more money than they had on iTunes. This caused their accounts to go into negative balance, and meant that you had to bring the balance back above zero before you could buy or update anything else.
