When Jesus said "Upon this rock I will build my church" he was speaking to Simon, his chief apostle. Simon's faith was the strongest (as you may know, Jesus chose him to walk on the water). At this moment, Jesus changed Simon's name to Peter, which literally means "rock". The verse is in Matthew's Gospel, see St. Matthew 16:17-19 in which Our Blessed Lord changes Simon's name to Peter (rock), a word that had never been used as a name before. This is common in The Bible when God is appointing someone for some great work (Abram->Abraham, Isaac, etc.) In changing his name, Our Blessed Lord appoints Peter and his successors as His Vicars, as the kings of Israel appointed their Vicars to run the kingdom. They spoke with the King's voice (read Isaiah 22:20-25 when the Lord God deposes Sobna and appoints Eliacim as his Vicar and gives him the keys to the Kingdom). It is at this instance that the papacy started, "pope" is the word that we use for the Vicar of Christ.
