In 1928 Sir Alexander Fleming observed that colonies of bacterium Staphylococcus aureus could be destroyed by the mold Penicillum notatum. At the time the importance of his discovery was not known. It wasn't until the 1940's when Howard Florey and Ernst Chain isolated the active ingredient and developed a powdery form of new medicine.

Penicillin is used to treat infections caused by bacteria, such as ear infections. So in other words, no, penicillin was NOT used to sterilized surgical instruments.
