Is there a button to activate the overdrive on the transmission. If the button is activated and not in overdrive mode the car will never shift into 4th gear.
If your transmission is not shifting into third gear and overdrive it could be a parking pawl. This prevents the transmission from rotating.
Could be low fluid,a bad sensor or transmission going bad.
Are you sure that it's third it's not shifting into, and not fourth? The transmission is designed to not permit shifts into fourth (overdrive) until the engine and transmission have warmed up a bit.
The Third Shift was created in 2004.
There are 3 Oxygen sensors on Lexus RX300. I have replaced the 2 forwards ones so far. The third one is Post- Catalytic converter and requires drivers seat removal to get at the electrical connector. It is also difficult to remove the O 2 sensor from the exhaust system because it is mounted into an exhaust hanger.
That car is equipped with a 4R70W electronic automatic transmission through the 2005 model year, and the Overdrive OFF switch is on the end of the shift lever. Push it in to disable Overdrive, and again to re-enable Overdrive. This feature is typically only used when towing or in very hilly or mountainous terrain that would cause the transmission to "hunt" between Third and Fourth gear a lot.
If this is an automatic transmission, which I am assuming it is it will not work the same as a standard. Only drive it in overdrive or 1st gear.

My 1984 corvette transmission is not shifting into overdrive. It shifts first thru third fine.
