Two prominent founding fathers, Benjamin Franklin and Thomas Jefferson, were "Children of the the Enlightenment". In other words, they were quite influenced by the teachings of people like John Locke, Rene Descartes, Baruch Spinoza, Immanuel Kant, Sir Isaac Newton, Voltaire, Jean Jacques Rousseau. The Declaration of Independence (1776), written by Jefferson, laid out the fundamental principles of America, in which he called for equality and liberty. Jefferson also advocated ending slavery and promoting religious tolerance, which was an immense part of the enlightened teachings.
I think you have it backwards, but that the United States democracy is based on the philosophy of Locke so the constitution agreed with Locke.

Figure it out. Don't be lazy
The Declaration of Independence is heavily based on John Locke's ideas in his "Second Treatise of Government". Thomas Jefferson's words: "life, liberty and the pursuit of happiness" were based on Locke's: "life, liberty and property". The first line of the Declaration that proclaims that "all men are created equal" and that it is our God given right to overthrow governments that aren't governing us properly comes directly from what Locke thought was man's "state of nature" and his idea of the "social contract". I'm not sure about any others.
John Locke

The Constitution uses ideas from philosophers from the Enlightenment era. For example, john Locke's ideas on securing life, How_did_the_enlightenment_ideas_influence_the_American_revolution_and_constitutionand property and Montesquieu's ideas on separation of powers and checks and balances.
John Locke was the first to think of the natural rigths a man or woman is born with. He influced Thomas Jefferson and a lot of his ideas influenced the US Constitution
