It is a reference to a 1970's TV series called "Hawaii 5-0" which was about a detective unit in Honolulu.
Added; Originally it was just applied to detectives or undercover police and/or their un-marked vehicles but eventually it came to be applied to the uniformed patrol units as well.

5-0 became a slang term for police after the hit show "Hawaii Five-O." The show was centered around a fictional division inside the State-Police of Hawaii and carried the same name as the television show ("Hawaii Five-0"). The division's name gets its origin from honoring the fact that Hawaii was the 50th state to become apart of the United States of America.
Well, both- sort of. There used to be a TV show about the Hawaii State Police, called Hawaii Five Oh- and it beame slang for the police. BTW, Hawaii does not have a state police department.
Technically, five-o refers to any police department. However, the term orginated because of the TV show Hawaii Five-O (the one from the 70s or 80s.)
But in the Uk they called them like bobs or something similar to that but it the Us you can called them PD,Police,Cops, or Five o we dont really know what the word Police camed from

Because of the old TV show "Hawai'i Five-O" - the 5-0 on the TV show just meant that Hawai'i was the 50th state. An alternate answer, is - a decade after the show, police in California drove cars with 5.0 engines. the engine size was displayed on the side (by the manufacturer - Ford). it became prison slang at this time. then tu PAC wrote a line something like "they threw him in the 5-0", in one of his songs, at that point white kids & the general population started using it. the Hawaii 5 0 reference is cute but it didn't start the useage.
The Police
well it now stands for the police. but it came from the original Hawaii 5-0 show Hawaii is the 50th state. hence 5-0 The 2010 series the 5-0 stands for Steve's football shirt on the back it said 50 and kono said let call our task force Five o.
They wore green uniforms, that's why they were called the green police.
