The biggest disadvantage of an electron microscope in the study of cells is that the cells must be fixed and prepared prior to viewing. Since many of the questions about cell function relate to the dynamic change of cells this is good reason to use other tools for this kind of research.

Advantages: You can see incredibly small particles with its intense magnifying power (up to about 20,000 times) Disadvantages: Using EM's, you cannot view objects/organisms in live color, or in a live state (shooting thousands of electrons at incredibly high speeds just doesn't agree with living systems)

Advantage of EM is that the resolusion power is very high so one can observe the structure of virus particle,varius cell organeles etc but main disadvantages are we can not observe live specimen,sample preparation is laborious and is very costly

The disadvantages of STM are listed as follows:

- It is slower compared to other techniques.

- It is mainly used to analyze conducting materials.

- It is more prone to mechanical vibrations and acoustic disturbances

- The best results from STM can be obtained only in vacuum conditions and hence it may not be the best tool to inspect ad analyse biological samples

They do not really allow you to see different tissues etc only the very small detials of the cells. There is no overview.

Samples are examined in a vacuum, so you can't examine either live subjects or vital processes. They also cost as much as a few hundred light scopes.
