Most of the rights enshrined in the Constitution of the United States are based in ideas of natural law, or the principle that in a state of nature, without government interference, man naturally engages in certain activities, and that government should not be permitted to interfere with this. For example, man naturally expresses his ideas and practices his religion. This activity is protected by the First Amendment . Man naturally protects himself, his family, and his property. This activity is (arguably) protected by the Second Amendment.

Other rights were included to prevent or limit the lengths to which government could get involved in the private lives of the people at all, or to prevent intrusive activities that prior governments had conducted. This is one reason why, under the Third Amendment, governments cannot quarter troops in private homes.

Finally, some rights are established purely to preserve some fundamental fairness in our system of government. For example, one has the right to confront his accuser, face to face, in a criminal trial, under the Sixth Amendment.

The national constitution also protects certain rights for the states, such as the right of each state to equal representation in the senate, the right to select its own method for choosing presidential electors, the right to call a constitutional convention, and the right to regulate the sale of alcohol. The purpose of these rights is to protect the states as independent sovereign entities which exist separate and apart from the national government.
