It depends on which journey is meant. Eyre's first major journey was his attempt to be the first to overland sheep from just west of Sydney to Adelaide. This journey began on 21 December 1837, departing from Limestone Plains. this particular expedition was unsuccessful. Eyre's next journey was to explore the countryside beyond the headwaters of the Hutt River, 100km north of Adelaide, which had not yet been settled. this journey commenced on 1 May 1839. Eyre's expedition to be the first to cross Australia from south to north departed Port Lincoln on 5 August 1839. His journey was thwarted by a series of salt lakes which Eyre believed to be one gigantic horseshoe-shaped inland sea, because by sheer bad luck, he was unable to find any of the breaks between the lakes. This was when he changed his plans and decided to try to be the first to cross from east to west. He returned south to Fowler's Bay and sent some of his men back to Adelaide, opting to take a smaller exploration party across the Nullarbor. This journey departed Fowler's Bay on 25 February 1841.

Edward Eyre explored primarily so his expeditions could benefit the new colony of South Australia. His expeditions were all undertaken for this purpose. For example, he wanted to be the first to drive cattle overland from Sydney to South Australia, a journey he attempted in 1837. To find an overland route for stock animals would benefit South Australia as it would mean easier transportation and trade of animals than having to take them by ship. He departed from Limestone Plains (today's site of Canberra), with over a thousand sheep and cattle. He travelled first to Melbourne, then tried to head directly west to Adelaide. Conditions were difficult, as it was a drought year, so he could not get through the mallee country of western Victoria. Eyre had to retrace his steps to the Murray River, and his first explorations were not successful. In 1839, Eyre left Adelaide to explore north in search of good pastureland that would allow settlers to move further out from Adelaide. He discovered excellent countryside just north of Adelaide and around today's Hutt River. It was on this journey that he discovered and named Mount Remarkable. Eyre finally arrived at the head of Spencer Gulf on 15 May 1839, where he discovered and named Depot Creek. Eyre left Adelaide in June 1840 to explore north towards the centre of Australia. On this journey, he wanted to bring the colony of South Australia to new prominence by achieving something new and noteworthy on its behalf. No-one else had mad a successful crossing from south to north. He did not get very far as he was blocked by salt lake after salt lake: each time he attempted to go around a salt lake, he found his way barred by yet another. This led to Eyre's theory that Adelaide was surrounded by a vast horseshoe-shaped salt lake. By sheer bad luck, Eyre was unable to locate any of the breaks between the numerous salt-lakes which encircled the area. However, Eyre was not prepared to return to Adelaide after an unsuccessful attempt. His great hero was the explorer Sturt, and Sturt had shown great faith in Eyre, helping him organise supplies and even giving him a specially autographed copy of his book of his own two expeditions. Sturt had been at the special send-off held in Adelaide in Eyre's honour. As a result, Eyre returned to Streaky Bay on the western coast of the Eyre Peninsula. From there he then travelled to Fowler's Bay, where he regrouped. He sent some of his men back to Adelaide, taking only those who volunteered to go with him. At this point, he chose to go west, which no-one had ever tried before. He finally reached Albany in June 1841 in an amazing journey of courage and endurance across the Nullarbor Plain.
