If the transmission is low on fluid then the pump that circulates fluid through the transmission and gives it the hydraulic pressure that allows it to work will cease to function. If you are low/out of trans fluid you should empty it completely and change the trans pan gasket and trans filter while you are at it (usually less than $15 if you are doing it yourself).
If you continue driving the vehicle without trans fluid it will break the transmission.
It will spill?
They have a sealed transmission you dont have to fill it. The fluid should be changed around 100,000 and it has to be done by a dealer.

1992 - 2002 Isuzu Troopers have the GM 4L30E transmission. There is no dipstick. There is no fill tube. The only way to add fluid or check the fluid level of the transmission is through the fill plug in the transmission pan. The transmission fill plug is situated above the fluid level of the "sump" area of the pan. Add fluid by pumping it into the fill plug and let any excess drain out. You'll know it's topped off when the fluid stops pouring out.
I always fill mine where the transmission dip stick goes in.

You put the Transmission Fluid in the Transmission (fill point transmission drip stick tube)

You fill the transmission fluid on the 1988 F250 diesel at the dipstick of the transmission. You will need a filler funnel in order to keep spillage to a minimum.
Remove the plug from the transmission fluid pan. Allow the transmission fluid to drain out. Replace the plug to its original position. Fill the transmission with new fluid. Remove the transmission filter by turning it to the left. Fill the filter with new fluid. Return the filter to its original position.
Remove the plug on the bottom of the transmission. Allow the transmission fluid to drain out. Replace the plug back to the bottom of the transmission. Fill the transmission with fluid.
A whining noise in the transmission is usually an indication the transmission fluid is low. Check the transmission fluid and fill it to the recommended level.
Use a funnel and pour the fluid down the dipstick tube making sure you dont overfill it.
