Doesn't "WANT" to work - it has no choice, it either works or it doesn't.
Is the sound setting correct on the iPod? Check that it hasn't been turned to silent.
Does it work connected to any other device? Try the earphone socket on your computer, or on a friend's MP3 player. If not, then the earphones are broken
Do other earphones work on the iPod? If not, then the iPod has developed a fault.

No, it will not make any damange.. it is completely safe to play your itouch while its charging in a computer or on the wall :) ... unless if you are syncing your itouch then dont play it.. only play it if its just charging

ive just had the same problem dont know what caused it but ive fixed it simply by removing the headphones and plugging my ipod into the laptop using the lead provided and letting the ipod syncronize. i left mine in then ejected it correctly and now the problem is fixed. i hope this works with yours.
