methylene blue test is basically a rapid method be determined the microbial load .

First of all it is require to sterilize all the equipments related to the experiment such as pipette, rubber cork, test tube etc. Then in each test tube methylene blue is added. After that those are incubated. then place them in water bath and after half an hour test tubes are taken out and filled with the sample. then again place them in water bath.Repeat this for a while. Note the reduction time of each test tubes. then you get the result of all sample you put in the test tubes

Methylene Blue is used as a stain to see various organelles clearly, especially the nucleus and effectively used for general microscopic study of plant (like onion skin or root hairs) and some animal tissues. The stain has a very low toxicity which keeps cell alive and as such is considered a "vital" stain.
