I've searched a bit and I couldn't find any direct route from Vancouver to Yukon ( or vice versa). The only way I found was: Vancouver - Prince George - Dawson Creek - Whitehorse.
One way trip by Greyhound:( all standard values, not the web only nor the refundable price)
Vancouver - Prince George = 92.00$ CAD
Prince George - Dawson Creek = 77.30 $ CAD
Dawson Creek - Whitehorse = 244.00 $ CAD
TOTAL: 413.00 $ CAD
I think it's cheaper to go by plane, which would cost around 700.00 $ CAD (round trip),by the Air North airline. ( the same flight in Air Canada would cost more than 1,000$ CAD)
For this winter(2013), there's a deal in Air North: 579$, round trip, including two nights in a hotel. It's a good price perhaps.( the link is the second below). Take a look at the websites below to better plan your trip and find the lowest prices. Perhaps you'll need to do some calculations, but it's worth to do so now, in order to save more money later.
